Semax

ACTH 4-10 analogue

Research Use Only

Russia's prescription nootropic. Developed for stroke recovery, adopted by biohackers for focus and memory.

Nootropic PeptideEvidence:

What is Semax?

Semax is one of the few peptides that's actually an approved medication — in Russia, where it's been prescribed since 2001 for stroke recovery, cognitive disorders, and ADHD. It's a synthetic analogue of ACTH(4-10), a fragment of the stress hormone ACTH, but without the hormonal side effects.

The nootropic community discovered Semax through Russian medical literature, and it's become one of the most popular cognitive enhancement peptides. Users report improved focus, reduced brain fog, enhanced verbal fluency, and better working memory — effects that are subtle but consistent.

How Does It Work?

Semax increases BDNF (Brain-Derived Neurotrophic Factor) — the protein responsible for growing new neurons and strengthening existing neural connections. It also modulates serotonin and dopamine metabolism, enhances acetylcholine signalling, and has anti-inflammatory effects in the brain.

Unlike stimulants, Semax doesn't provide a 'buzz' — it works by improving the underlying neural infrastructure. Think of it as fertiliser for your brain rather than a Red Bull.

What Does The Research Say?

Evidence Quality:(3/5)

Moderate evidence. Some human data, mostly animal studies.

Russian clinical trials support its use for stroke recovery, cognitive impairment, and optic nerve disorders. It's been studied in thousands of patients. Western research is limited but growing — several studies confirm BDNF elevation and neuroprotective effects.

The evidence quality is moderate: strong in Russian literature, limited but supportive in Western studies. The main limitation is the lack of large Western double-blind trials.

Reported Dosages

These are dosages reported in research literature and community reports. They are NOT medical recommendations. Always consult a healthcare professional.

Russian prescription: 200-600mcg intranasally, 2-3 times daily.

Nootropic community reports: 200-600mcg intranasally or subcutaneously daily.

N-Acetyl Semax Amidate (NASA) is a modified version with reportedly longer duration.

These are research dosages, not medical recommendations.

Side Effects & Risks

Generally well-tolerated in clinical use. Occasional reports of: mild headache, nasal irritation (intranasal route), transient hair shedding (rare), irritability at high doses. No significant adverse effects reported in Russian clinical studies.
