01 What is MK-677?
MK-677 is not technically a peptide — it's a non-peptide growth hormone secretagogue that mimics the action of ghrelin (your hunger hormone). What makes it interesting is that it's orally bioavailable, meaning you swallow a pill instead of injecting. It stimulates your pituitary gland to release more growth hormone (GH) and IGF-1, which is why the bodybuilding and anti-aging communities are obsessed with it. The appeal is obvious: growth hormone therapy costs thousands per year and requires daily injections. MK-677 offers a fraction of the GH boost for a fraction of the price, with none of the needles. The trade-off? It's not as potent, it makes you ravenously hungry (ghrelin agonist, remember), and the long-term safety data is limited.
02 How Does It Work?
MK-677 binds to ghrelin receptors (GHSR) in the brain, triggering a pulse of growth hormone from the pituitary. Unlike exogenous GH injections, this maintains your body's natural pulsatile GH release pattern. It also increases IGF-1 levels by 40-80% in most studies, which drives the muscle growth, recovery, and anti-aging effects. Importantly, MK-677 doesn't suppress your body's natural GH production — it enhances it. This is a key advantage over synthetic GH injections, which can cause your pituitary to get lazy.
03 What Does The Research Say?
Moderate evidence. Some human data, mostly animal studies.
Multiple human clinical trials exist, which is more than most research compounds can claim. Studies show sustained GH and IGF-1 elevation over months of use, improved sleep quality (deeper stage 3/4 sleep), increased lean mass in elderly subjects, and improved bone mineral density. However, metabolic effects are a concern: MK-677 can increase fasting blood glucose and reduce insulin sensitivity, particularly at higher doses. One 2-year study in elderly patients was discontinued partly due to these metabolic effects.
04 Reported Dosages
Research literature dosages only. NOT medical recommendations. Always consult a healthcare professional.
Research studies typically use 10-25mg daily, taken orally. Most commonly 25mg once daily, usually at night (to align with natural GH pulse during sleep). Some users prefer lower doses of 10-15mg to minimize hunger and metabolic side effects. These are research dosages, not medical recommendations.
05 Side Effects & Risks
Increased appetite (significant — this is a ghrelin agonist), water retention, lethargy in the first few weeks, potential blood sugar elevation, numbness/tingling in extremities (from GH effects). Long-term concerns include insulin resistance and potential IGF-1-related risks.
